1st English colony because it was established in 1607, before Massachusetts in 1620, that succeeded. There was an attempt in the late 1580's to establish an English colony called Roanoke in the outer banks region of what is now North Carolina.

The Spanish were colonizing before that, but 'colonizing' might not be the correct term for Spain's 1st involvement in the New World. Since 1492 Columbus expeditions to the west, the Spanish were sending 'explorers' / conquerors to Central America, especially Mexico. St. Augustine, Fl., was established in 1565, so it was before Virginia.
