Not an easy question to answer. Strongest often means largest. Here's a brief sample of why answering is a problem:

• Largest land artillery piece in the US arsenal - Atomic Annie (currently on display at the US Army Artillery Museum at Ft. Sill) the M65, weighed 83.3 tons towed, 280 mm. Test fired at Frenchman Flats, Nevada Test Site, 25 May 1953, 0830 local. The W9, 15kT projectile (the only nuclear artillery round ever tested) traveled seven miles. The unclassified effective range was 20 miles, but rumor has it the cannon was able to reach half again as far.
• Largest naval artillery piece in the US arsenal - Mark 7 16" guns fitted to the Iowa class battleships.
• World's largest working revolver - replica Remington 1859 built by Ryszard Tobys from Poland. Weighs 99.2 pounds, 4' 1.5" long, 15.75" high. Cylinder holds six 28mm bullets.
• Largest sniper rifle - Barrett M107 .50 shoulder fired rifle.
• World's largest centerfire rifle - SKS .950 JDJ. Fires a 2400 grain ball with 230 grains of powder, with the kinetic energy of a WW2 Panzer round.
• World's largest artillery piece - the Schwerer Gustav Super-Heavy Railway Gun fielded by the Wehrmacht. Weighed 1,350 tonnes, 47.3 meters long, fired 81 centimeter 4,800 to 7,100 kg HE and armor piercing (respectively) rounds. Range, 48 km.
• World's largest handheld pistol - S&W Model 500, .50 caliber revolver.
• World's longest range gun - (Nomenclature unknown) on 10 December 2010, the US Navy test fired the largest rail gun (projectiles launched by electromagnet as opposed to conventional powder charge) ever fired. The size of the projectile was not reported, but it exceeded Mach 7 (+2382.03 meters/second) and traveled over 100 miles.

In addition, Russia built the 890 mm Tsar Cannon in 1586, two 910 mm shore guns were built by the UK (Mallet's Mortar) in 1857 and the US (Little David) in 1945, the Russian piece was a bombard, the UK and US pieces were mortars, generally referred to by these names or shoreguns and not the less specific guns.

I think the most powerful standard production handgun is the Smith and Wesson 500, which fires a 50 caliber bullet.

There is a non standard production handgun called the 600 nitro express that I think is more powerful.
