It was a campaign slogan for James Polk in the presidential election of 1844. It referred to the upper latitude boundary line of the joint controlled ( British/ American) area north of Oregon: 54 degrees forty minutes north. Polk campaigned on the platform of the US getting full control of the area....in other words "let's get control all the way to the 54-40 line, or fight for it"
