What are restriction enzymes?

Restriction enzymes (also known as restriction endonucleases) are proteins which cut DNA up at specific sequences in the genome. For example, the commonly used restriction endonuclease EcoRI recognizes every point in DNA with the sequence GAATTC, and cuts at the point between the Guanine and Adenine. Interestingly, the recognition sequences for most restriction endonucleases are genetic palindromes, e.g., the sequence reads exactly the same backwards on the complementary strand. In the case of EcoRI, the two complementary DNA strands for the recognition sequence are: 5'--GAATTC ---3'3'--CTTAAG--5'


Why can a paternity test only disprove paternity?

Most paternity tests are based on comparisons of DNA. DNA evidence can only exclude suspects. DNA sequencing is used because there are so many different sequences you could have at any one place, the odds of any 2 people, except identical twins, having the same DNA is nearly impossible. However, if people are very closely related (like father-son), they will share a number of common sequences on a DNA test. We'll say 5 out of 10. Though it's usually VERY unlikely, 2 completely unrelated people can still share sequences in certain places. This is why it can only legally disprove paternity cases. If 2 people shared no sequences, they couldn't be related. That's not to say, however, that 2 people who share a number of similarities are related.
