I'm not sure about today's marking substance, but for many years, in the old days, it was nothing more than POWDERED CHALK, which I think is Calcium Carbonate. Today, some are using synthetic materials, or even spray paint.

In the early years of American football, the field had vertical lines as well as horizontal lines, creating the "gridiron" (or checkerboard) effect. The rule was that each play would begin in the square at which the previous play ended. The vertical lines were replaced by hash marks sometime around 1910-11.

Why the lines in all sports field are marked five centimeters?

The answer here lies not with the field but in the applicator of the paint used for the field lines. 5cm is really the arbitrary width of sprayer they use on that strolling painter machine. The reason it's centimeters and not inches is that those parts were probably made in Canada.
