The origin for transmutation circles came from the anime FullMetal Alchemist. Transmutation circles are formed using the symbols and keys of real life alchemy used during the Middle Ages. Their meanings and the meaning interpreted into the show are very similiar, but that is only done to encourage the belief of it's existence. Creating works that can be linked to our past or everyday lives tends to make us believe in it's existence. So, to answer your question, transmutation circle do not work. They were created to endorse a now famous Japanese anime. and that is all they will ever be.

no i don't think so but there are only three people in the full metal alchemist world that can do alchemy without a transmutation circle and the three that can use alchemy without a transmutation circle are the two elric brothers and their alchemy teacher.

However, they were creative with how they did this. Mustang wore gloves that had a transmutation circle on them, as did Armstrong (though his "gloves" were more like steel gauntlets). Scar, an Ishbalan who despises alchemy, actually uses his right arm, which has an incomplete transmutation circle of sorts tattooed on. Edward of course doesn't have to use a circle because of the knowledge he gained while inside The Gate (the place he went to when he transmuted his brother and dead mother).

In the world of Fullmetal Alchemist, an alchemist can preform transmutations without a circle if they have seen 'the truth' like Edward Elric has. 'The truth' is often seen when at the Gate, which usually appears when a human transmutation occurs. Other alchemists that can transmute with a circle are Izumi Curtis and Alphonse Elric.
