Reagent Blank : Take reagent and add deionised water (in place of sample to be tested). Now measure the OD at specific wavelength --> this OD is your reagent blank. Substract this OD from your test result (with sample) to avoid any false +ve effect due to colour of reagents itself.

Sample Blank : Take sample and measure the OD without adding reagents --> this OD is your sample blank. Substract this OD from your test result to avoid any false +ve effect due to colour and turbidity of sample itself. As it is the fact that colour and turbidity of each sample would vary from one to another.

So now it is clear that Reagent blank is used to avoid bias due to colour of reagents and Sample blank is used to avoid bias due to sample itself.

If you want to read the absorbance at 595nm of a protein solution mixed with the colorizing solution called Bradford reagent what is a suitable reagent blank?

A suitable reagent blank for measuring the absorbance of a protein solution mixed with Bradford reagent at 595nm would be a blank containing all components of the reaction except the protein sample, such as water or buffer mixed with the Bradford reagent. This blank will account for any background absorbance contributed by the reagent itself, allowing for a more accurate measurement of the protein concentration.

What is a reagent blank?

A reagent blank is a sample containing all the reagents of a particular test except the substance being tested. It is used to account for any background interference or contamination that might affect the test results. This helps to ensure that the measured signal is due solely to the presence of the substance being tested.

Why do we add water to reagent blank?

We add water to the reagent blank to ensure that any impurities or contaminants in the water or the other reagents used will be accounted for in subsequent measurements, allowing for a more accurate determination of the analyte concentration. This helps to correct for any background signal or interference that may affect the final results.

What is the difference between a blank monthly calendar and a regular calendar?

The difference between a blank monthly calendar and a regular calendar is that there are days of the weeks with numbers written on a regular calendar. A blank monthly calendar has nothing written on it, which you can fill in yourself.

What is the purpose of the blank?

Blank Sample in Spectrophotometry is used to measure the absorbance of light without sample. It is subtracted from the total absorbance for measurement of Absorbance from a sample's absorbance.

Why does blank titration shows the higher value compared to sample titration in iodine value?

In blank titration, no sample is present to react with the iodine solution, leading to an apparent excess of iodine. This can result in a higher value as all the iodine being counted towards the blank. In sample titration, the sample reacts with the iodine, leading to a lower amount of iodine available to react, resulting in a lower value compared to the blank titration.

Why do you use a reagent blank and not just distilled water to zero the spectrophotometer?

AS with any accurate machinery or scientific instrument there is a tendancy for them to get out of calibration over time, and after use. These products must meet specific requirements, codes, and standards. The purpose of "Zeroing" (or calibrating) is to make sure these items are brought back into "Zero" or "calibration" so that they can be as accurate as possible pertaining to the specifications that are required by the governing requirements, etc.. Why is a blank solution used in morhs method?

A blank solution is used in the Mohs hardness test to serve as a point of reference for comparison. By observing how the blank solution interacts with the unknown mineral sample, the tester can accurately assess the hardness of the sample by determining if it scratches, is scratched by, or is comparable in hardness to the blank solution.
