First, write it out.

3x+4y=16

Now, I was taught to find the y intercept by using slope intercept form, so pardon me if I am donig it differently than the way you are used to :p

You want to get the y on one side, alone, so subtract the x.

3x+4y=16

-3x ......-3x

4y=-3x+16

Now, you want the y to have no coefficient, therefore you divide EVERYTHING by 4.

4y=-3x+16

/4 ..../4 ....../4

y=-3/4x+4

Your y intercept is the lone number, hence "4".
