Slope is the change of your y variable divided by your change in the x variable.

So let's say you start at (3,0) and walk to (6,12)

Then your slope (generally called "m") is

m = (y2 - y1)/(x2 - x1) = (12 - 0)/(6 - 3) = 12/3 = 4

So your slope is 4. That means for every one you move right, you move four up.
